Uridine is a pyrimidine nucleoside that plays a key role in brain phospholipid synthesis, synaptic plasticity, and dopamine modulation. It is studied for its potential to enhance memory, mood, and neuronal membrane integrity.
Uridine is one of the four nucleosides in RNA and serves as a building block for phosphatidylcholine (PC), the most abundant phospholipid in neuronal cell membranes. When taken orally as uridine monophosphate (UMP), it crosses the blood-brain barrier and is converted to cytidine-5'-triphosphate (CTP), which is rate-limiting in the Kennedy pathway of phospholipid synthesis. Uridine synergizes with omega-3 fatty acids (DHA) and choline to promote synaptogenesis, dendritic spine formation, and neuronal membrane repair. Research by Wurtman and colleagues at MIT has demonstrated that this tripartite combination increases synaptic protein levels and neurite outgrowth. Uridine also modulates dopaminergic neurotransmission, with preclinical evidence showing increased dopamine release and receptor expression. It is found naturally in breast milk, beer, and organ meats, and is available as a dietary supplement in the form of uridine monophosphate.
Uridine is a rate-limiting precursor in the Kennedy pathway for phosphatidylcholine synthesis. Combined with DHA and choline, it significantly increases synapse formation and dendritic spine density in animal models.
